So I noticed a lake that has no access to other lakes and has a once-a-week permit entry. The permit is Saturday to Saturday. What if I want to go in a Friday and out on a Sunday, do I need to reserve two weeks? There is only one open campsite on the lake.

You must be looking at Bog Lake. Here is what the permit reservation site says:

"The Bog Lake Entry Point only has one campsite available. Permit users are only able to enter on Saturdays, but may camp up to 14 days at one designated campsite per USFS regulations."

So you are out of luck if you want to enter on a Friday. Likewise, you might be out of luck if the person from the week before decided to stay 14 days - or 8.

Perfect- that's what I wanted to know! So Bog Lake has 3-4 campsites, only one open. Does that mean you cannot camp at a closed site or just that it's not being maintained?

You cannot camp at them. The Pagami Fire destroyed them, and they are closed so they can recover. I have no idea what you would do if someone is at the only campsite - ask to stay with them? Not sure what the Forest Service expects you to do.
